Efficient Re-Differentiation of Rice (Oryza sativa L.) Callus Induced by Exogenous Hormones Indole-3-Butytric Acid and Thidiazuron in Tissue Culture

摘要: Exogenous hormones play a significant role in the process of plant growth and tissue culture. The suitable hormones with reasonable ratio of concentration can effectively promote the proliferation and differentiation of rice callus. In this study, we explored the effects of 2,4-dichlorophenoxy acetic acid (2,4-D), indole-3-acetic acid (IAA), indole-3-butytric acid (IBA) and 6-benzylaminopurine (6-BA), kinetin (KT) and thidiazuron (TDZ) on proliferation and re-differentiation of rice callus, and investigated their differences. Then, we chose IBA and TDZ for concentration ratio experiment to optimize hormone concentration for re-differentiation of rice callus. The results showed that 0.4 mg/L TDZ effectively promoted the re-differentiation of rice callus, and 2 mg/L IBA further improved the proliferation rate of rice callus. It has been well-known that appropriate auxin and cytokinin had a mutually reinforcing effect, so when the medium was added 0.5 mg/L TDZ and 2.5 mg/L IBA, the differentiation efficiency of rice callus could reach the optimized level.
